2'-deoxynucleoside 5'-phosphate N-hydrolase 1 (DNPH1) is an enzyme that catalyzes the hydrolysis of glycosidic bonds in deoxyribonucleotides, specifically detoxifying 5-hydroxymethyl-dUMP (hmdUMP) to prevent its misincorporation into DNA. By modulating the nucleotide pool, DNPH1 protects genomic integrity and contributes to cell proliferation regulation. Upregulated by c-Myc, it supports the metabolic demands of rapidly dividing cells. Inhibition of DNPH1 sensitizes cancer cells to PARP inhibitors and chemotherapy by accumulating cytotoxic nucleotides. |
