| 背景信息 | Perilipin 1 (Perilipin-1) is a primary lipid droplet-coating protein predominantly expressed in adipocytes, where it serves as a dynamic regulator of lipid metabolism. Under basal conditions, it protects lipid droplets from premature degradation by sequestering lipases such as hormone-sensitive lipase (HSL) and adipose triglyceride lipase (ATGL). Upon PKA-mediated phosphorylation in response to lipolytic stimuli, Perilipin-1 undergoes a conformational change that recruits these lipases to the droplet surface, facilitating triglyceride hydrolysis. It also interacts with specific phospholipids and neutral lipids to maintain droplet stability. Clinically, reduced expression of Perilipin-1 is associated with obesity-related lipodystrophy and insulin resistance, while genetic polymorphisms in the PLIN1 gene are linked to metabolic syndrome. |
