Geminin DNA replication inhibitor (GMNN) is a nuclear protein that strictly regulates DNA replication and cell cycle progression. It functions by binding to the replication factor Cdt1, preventing the re-loading of the MCM complex onto chromatin and ensuring that DNA replication occurs only once per cell cycle. Geminin levels fluctuate during the cell cycle, accumulating in S and G2 phases and being degraded during mitosis. Beyond replication control, Geminin influences cell lineage commitment and neural differentiation through interactions with chromatin remodeling complexes. Its overexpression is linked to various cancers, acting as an oncogenic factor, while germline mutations are associated with Meier-Gorlin syndrome, highlighting its role in development and genomic stability. |
