| 背景信息 | Argininosuccinate synthase 1 (ASS1) is a rate-limiting enzyme of the urea cycle that catalyzes the synthesis of argininosuccinate from citrulline and aspartate. This reaction is critical for the detoxification of ammonia and the endogenous production of arginine. ASS1 expression is regulated by nutrient availability and circadian rhythms. Deficiency in ASS1 causes citrullinemia type I, a severe metabolic disorder characterized by hyperammonemia and neurological impairment. In oncology, ASS1 expression is frequently silenced in various tumors, rendering them auxotrophic for arginine. This metabolic vulnerability is exploited therapeutically using arginine-depleting agents like pegylated arginine deiminase. |
