Interleukin 11 (IL-11) is a cytokine that was initially thought to be important for hematopoiesis and platelet production, but more recent studies have revealed a more complex role for IL-11 in disease pathobiology. While IL-11 was previously considered to have anti-inflammatory and regenerative effects, newer research has shown that IL-11 is actually pro-fibrotic, pro-inflammatory, and anti-regenerative . IL-11 signaling pathways converge to cause mesenchymal transitions and a pro-inflammatory state across different cell types, leading to stromal inflammation, fibrosis, loss of specialized cellular functions, and organ failure . IL-11 is upregulated in response to various disease factors as an "alarmin-like" response, and its overexpression has been associated with a number of diseases, including certain cancers, fibrotic diseases, and vascular diseases . As a result, IL-11 is now viewed as a potential therapeutic target, with several clinical trials investigating anti-IL-11 therapies for fibrotic diseases . |
