Interleukin 6 cytokine family signal transducer (IL6ST), also known as gp130, is the shared signaling subunit for the interleukin-6 (IL-6) family of cytokines. It mediates diverse physiological processes, including immune responses, hematopoiesis, and bone metabolism. The receptor consists of an extracellular region with cytokine-binding domains and a cytoplasmic tail that, while lacking intrinsic kinase activity, associates with JAK/STAT, Ras/Raf, and Src-family kinases upon ligand-induced homodimerization. Signaling is initiated when IL-6 binds to IL-6Rα, followed by the recruitment of gp130 to form a high-affinity hexameric complex. Clinically, dysregulation of the IL-6/IL6ST axis is central to the pathogenesis of rheumatoid arthritis, Castleman disease, and cytokine release syndrome. Therapeutic inhibitors targeting this axis are widely used to manage these inflammatory conditions. |
