Kinase suppressor of ras 2 (KSR2) is a large cytosolic scaffold protein that organizes the RAF–MEK–ERK MAPK signaling cascade. Although it possesses low intrinsic kinase activity, it acts as a location-regulated allosteric activator by binding BRAF and MEK1 to facilitate their phosphorylation. Predominantly expressed in the brain and metabolically active tissues, KSR2 also interacts with AMP-activated protein kinase (AMPK) to link Ras/MAPK signaling with energy-sensing pathways, thereby regulating feeding behavior, substrate utilization, and adaptive thermogenesis. Loss-of-function variants in KSR2 are strongly associated with severe early-onset obesity, hyperphagia, reduced basal metabolic rate, and marked insulin resistance, establishing it as a critical regulator of systemic energy homeostasis. |
