WW and C2 domain containing 1 (WWC1), also known as KIBRA, is a cytoplasmic scaffolding protein that integrates multiple signaling pathways via its WW, C2, and PDZ-binding domains. It acts as a critical regulator of the Hippo signaling pathway by promoting LATS kinase activity and suppressing YAP-dependent transcription, thereby controlling cell proliferation and organ size. In the nervous system, WWC1 modulates synaptic plasticity and AMPA receptor trafficking, processes underlying learning and memory. Genetic variations in WWC1 are associated with human memory performance and cognitive traits. Furthermore, its involvement in dendritic spinogenesis and protein trafficking links it to neurodegenerative conditions, including Alzheimer’s disease. |
