Delta/Notch like EGF Repeat Containing (DNER) is a single-pass type I transmembrane protein characterized by multiple epidermal growth factor (EGF)-like repeats, which mediate cell-cell interactions. Predominantly expressed in the brain, DNER functions as a Notch ligand, regulating neuron-glia communication and neuronal differentiation. It contains 12 EGF-like domains and a Delta/Serrate/LAG-2 (DSL) motif, enabling interactions with Notch receptors to activate downstream signaling pathways critical for cerebellar development and synaptic plasticity. DNER is implicated in astrocyte-mediated regulation of Purkinje cell morphology and function, underscoring its role in central nervous system homeostasis. Dysregulation of DNER is associated with neurological disorders, including autosomal recessive spinocerebellar ataxia type 14 (SCAR14), characterized by cerebellar atrophy, gait abnormalities, and intellectual disability. Additionally, altered DNER expression has been observed in certain cancers, such as breast cancer and glioblastoma, where it may influence tumor progression via Notch-dependent mechanisms. |
