Protein phosphatase 1 regulatory inhibitor subunit 14A (PPP1R14A), also known as CPI-17, is a protein that plays a crucial role in regulating smooth muscle contraction and has been implicated in various cancers. It acts as a phosphorylation-dependent inhibitor of smooth muscle myosin phosphatase, with over 1000-fold higher inhibitory activity when phosphorylated, creating a molecular switch for regulating the phosphorylation state of PPP1CA substrate and smooth muscle contraction. In cancer, PPP1R14A has been associated with tumorigenesis and progression, particularly in sporadic vestibular glioma, human melanoma, and schwannoma. High expression of PPP1R14A in most cases is linked to poor overall survival, disease-specific survival, and progress-free interval across patients with various malignant tumors, including adrenocortical carcinoma and bladder urothelial carcinoma. |
