Nucleosome assembly protein 1 like 3 (NAP1L3) is a nuclear histone chaperone belonging to the nucleosome assembly protein (NAP) family. It plays a critical role in chromatin remodeling and transcriptional regulation by binding core histones and facilitating nucleosome assembly during DNA replication and mitosis. NAP1L3 is particularly essential in hematopoietic stem cells, where it maintains the stem cell pool and regulates proliferation and differentiation. Mechanistically, it influences the expression of key regulatory genes, including MYC, E2F targets, and HOXA clusters. Depletion of NAP1L3 leads to G0 cell-cycle arrest and apoptosis, highlighting its importance in cell-cycle control and genomic stability. |
