Ankyrin repeat family A member 2 (ANKRA2) is a scaffold protein characterized by multiple ankyrin repeats that facilitate diverse protein-protein interactions, including binding to histone deacetylases and low-density lipoprotein particle receptors. It localizes to the cytosol and plasma membrane, where it serves as a key component of the 3M complex alongside CCDC8, CUL7, and OBSL1. Within this complex, ANKRA2 mediates interactions with HDAC4 and HDAC5, playing a regulatory role in protein complex assembly and the function of LRP2/megalin. Clinically, mutations in ANKRA2 are associated with Immunodeficiency 31B, characterized by recurrent infections due to T-cell and antibody deficiencies, as well as Familial Hypercholesterolemia 4, which involves elevated LDL cholesterol levels and increased cardiovascular risk resulting from receptor dysregulation. |
