Acid phosphatase 6, lysophosphatidic (ACP6) is a mitochondrial phosphomonoesterase that specifically hydrolyzes lysophosphatidic acid (LPA) into monoacylglycerol and free phosphate. It exhibits a preference for LPA substrates with medium-length fatty acid chains and functions optimally at low pH. By regulating the levels of LPA, ACP6 modulates downstream signaling through G protein-coupled receptors that control cell growth, proliferation, and wound healing. The enzyme's structure includes a hydrophilic water channel and an active site stabilized by specific arginine and histidine residues. Clinically, ACP6 is frequently downregulated in ovarian cancer, often as a result of gain-of-function p53 mutations. This loss of ACP6 leads to elevated LPA levels, which drive focal adhesion signaling, increased cell motility, and metastasis, marking it as a significant factor in tumor progression. |
