DNA methyltransferase 3 beta (Dnmt3b) is an enzyme essential for establishing de novo DNA methylation patterns during mammalian embryonic development. It catalyzes the transfer of methyl groups to cytosine residues in CpG dinucleotides, a process critical for gene silencing and genomic stability. DNMT3B contains a PWWP domain that guides it to specific chromatin regions by binding H3K36me3 marks. It functions cooperatively with DNMT3A and DNMT3L. Pathogenic mutations in DNMT3B cause immunodeficiency-centromere instability-facial anomalies (ICF) syndrome, a disorder characterized by hypomethylation of pericentromeric heterochromatin, immune defects, and developmental abnormalities. |
