Testin LIM domain protein (TES) is a cytoskeletal scaffold protein comprising an N-terminal PET domain and three C-terminal LIM zinc-finger domains. It localizes to focal adhesions and actin stress fibers, where it regulates cell spreading, motility, and cytoskeletal reorganization. Functioning as a tumor suppressor, TES negatively regulates cell proliferation and is frequently silenced in malignancies—including ovarian, breast, and gastric cancers—via promoter hypermethylation and loss of heterozygosity. Reduced TES expression correlates with epithelial-mesenchymal transition (EMT) and poor prognosis, suggesting its potential utility as a biomarker for cancer progression and therapeutic response. |
