Stomatin like 2 (STOML2) is a mitochondrial inner membrane protein belonging to the SPFH superfamily that plays a vital role in mitochondrial biogenesis and function. It binds cardiolipin to form specialized membrane microdomains that recruit prohibitins, optimizing the assembly of the respiratory chain complex and regulating calcium homeostasis. STOML2 also anchors the SPY protease complex for mitochondrial quality control. Beyond the mitochondria, it localizes to plasma membrane rafts in T cells, where it facilitates T cell receptor signaling and activation. Clinically, STOML2 is frequently overexpressed in various malignancies, including hepatocellular carcinoma and breast cancer, where it promotes cell proliferation, resistance to apoptosis, and metastasis, often correlating with a poor clinical prognosis. |
