| 背景信息 | Photoreceptor cilium actin regulator (PCARE) is a ciliary protein essential for the maintenance of photoreceptor cells and visual function. It regulates actin dynamics within the connecting cilium by interacting with WASF3 to activate the ARP2/3 complex, thereby promoting the F-actin polymerization required for outer segment disk morphogenesis. PCARE contains specific domains for ciliary trafficking, actin binding via a WH2 motif, and nuclear localization. It also interacts with components of the centrosome and microtubule-associated motors, indicating a broad role in ciliary structural organization. Clinically, mutations in the PCARE gene are well-established causes of autosomal recessive ciliopathies, specifically retinitis pigmentosa 54 and cone-rod dystrophy. These conditions lead to progressive photoreceptor degeneration and vision loss, underscoring the protein's vital role in the structural integrity and functional survival of retinal neurons. |
