| 背景信息 | Aconitate decarboxylase 1 (ACOD1), also known as IRG1, is a mitochondrial enzyme that catalyzes the decarboxylation of cis-aconitate to produce itaconate. This reaction is a cornerstone of immunometabolism, linking the tricarboxylic acid cycle to innate immune defense. Inducibly expressed in immune cells via TLR and NF-kappaB pathways, ACOD1-derived itaconate exerts broad anti-inflammatory and antimicrobial effects by inhibiting succinate dehydrogenase, modifying proteins via alkylation, and upregulating antioxidant defenses. Dysregulation of ACOD1 is implicated in tumor progression, neurodegeneration, and immune paralysis, highlighting its pivotal role in balancing metabolic flux during inflammatory responses and pathogen defense. |
