| 背景信息 | Forkhead box protein P3 (FOXP3) is a master regulator of regulatory T cell (Treg) development and function, playing a crucial role in maintaining immune homeostasis and self-tolerance. It contains several functional domains, including a proline-rich N-terminal domain, a zinc-finger and leucine-zipper domain, and a C-terminal forkhead domain responsible for DNA binding. FOXP3 acts as both a transcriptional activator and repressor, interacting with various proteins to modulate gene expression in Tregs. It is essential for the stability and suppressive function of Tregs, which suppress excessive immune responses and prevent autoimmunity. Mutations or dysregulation of FOXP3 can lead to severe autoimmune diseases like IPEX syndrome, characterized by multi-organ inflammation and autoantibody production. Additionally, altered FOXP3 expression has been linked to cancer, where it may influence tumor immunity and progression. |
