| 背景信息 | (SIRPB1) is a type I transmembrane glycoprotein of the immunoglobulin superfamily, primarily expressed on myeloid cells including monocytes, macrophages, dendritic cells, and neutrophils. Unlike inhibitory SIRP family members, SIRPB1 lacks cytoplasmic ITIM motifs and instead functions as an activating receptor. Its transmembrane segment associates with the ITAM-bearing adaptor protein TYROBP (DAP12). Upon ligand engagement, SIRPB1 triggers DAP12 phosphorylation and the recruitment of SYK-family kinases, which promotes phagocytosis, cell adhesion, migration, and the production of pro-inflammatory cytokines. Through these mechanisms, SIRPB1 modulates innate immune responses and the tumor microenvironment. Altered SIRPB1 signaling is implicated in various conditions, including glioma, autoimmune disorders, and chronic inflammatory diseases. |
