| 背景信息 | Matrilin 2 (MATN2) is a large, multifunctional adapter protein of the extracellular matrix (ECM) that contains five von Willebrand factor A domains. It is essential for the assembly of filamentous network structures, achieved through interactions with collagens, fibrillin-2, fibronectin, and laminin-nidogen complexes. MATN2 exists in multiple splice variants that influence its oligomeric structure and biological activity. The protein is critical for tissue repair and regeneration, including skeletal muscle, peripheral nerves, and liver tissue, by modulating TGF-β and BMP-7 signaling pathways. Clinically, MATN2 is involved in the reorganization of tissue architecture during liver cirrhosis and hepatocellular carcinoma. It also exhibits tumor-suppressive properties in ovarian cancer by regulating the PTEN/PI3K/AKT pathway. Furthermore, its dysregulation is linked to impaired wound healing and altered keratinocyte-fibroblast dynamics. |
