| 背景信息 | Von Willebrand factor A domain containing 5A (VWA5A), also known as BCSC-1 or LOH11CR2A, is a protein-coding gene located on chromosome 11 that encodes a protein primarily found in the nucleoplasm. The VWA5A protein features a von Willebrand factor A (vWF) domain, characteristic of extracellular matrix proteins involved in cell adhesion and signaling, with potential roles in protein-protein interactions and structural integrity; it exhibits paralogs like VWA5B1 and shows expression variations across tissues, often downregulated in aggressive cancers. Functionally, VWA5A acts as a tumor suppressor, where its lower expression correlates with enhanced cell proliferation, invasion, and migration in luminal and triple-negative breast cancer (TNBC) cell lines, but inversely in HER2 types; genomic deletions near CHEK1 further amplify instability upon VWA5A loss. In diseases, reduced VWA5A serves as a biomarker for early metastasis in breast cancer, predicting shorter metastasis-free intervals via proteomics and machine learning analyses, and associates with spinocerebellar ataxia 14 and mirror agnosia. |
