| 背景信息 | Delta like canonical Notch ligand 1 (DLL1) is a type I transmembrane protein that serves as a primary ligand for Notch receptors. Its structure includes an N-terminal MNNL domain, a DSL domain, and eight EGF-like repeats, which are essential for calcium-dependent binding and the subsequent activation of Notch signaling. Upon binding, DLL1 triggers receptor proteolysis and the nuclear translocation of the Notch intracellular domain. DLL1 is a critical regulator of neural stem cell quiescence and acts as an asymmetric fate determinant during cell division. Structurally, it possesses a rigid proline-rich region that reduces its binding affinity for Notch1 compared to other ligands like DLL4. The extracellular domain also contains a hydrophobic patch for receptor interaction and a DOS motif in the EGF repeats that is vital for signaling efficiency. |
