| 背景信息 | Neurocan (Neurocan) is a secreted extracellular matrix chondroitin sulfate proteoglycan of the lectican family, primarily expressed in the developing and remodeling central nervous system. It possesses a characteristic structure including an N-terminal hyaluronan-binding domain and a C-terminal lectin-like domain, allowing it to interact with hyaluronan, tenascin-C, and various cell-adhesion molecules like NCAM and L1. Through these interactions, neurocan modulates neuronal adhesion, cell migration, and neurite outgrowth. It is notably upregulated following nervous system injury, where it acts as a potent inhibitor of axon regeneration. In a clinical context, neurocan overexpression is associated with increased malignancy in neuroblastoma, where it promotes tumor sphere formation, stemness-associated gene expression, and the maintenance of the cancer stem cell niche. |
