| 背景信息 | Integrin subunit beta 1 binding protein 1 (ITGB1BP1) is a cytoplasmic adaptor protein that specifically regulates integrin-mediated cell-matrix interactions by binding to the β1 integrin cytoplasmic tail. It utilizes a C-terminal phosphotyrosine-binding (PTB) domain to compete with other activators like talin and kindlins, thereby negatively regulating the activation and clustering of α5β1 integrin. Through this mechanism, ITGB1BP1 modulates focal adhesion assembly, cell spreading, migration, and angiogenesis. It also integrates mechanical cues with signaling pathways such as MAPK/ERK and Notch to fine-tune cellular proliferation. Pathogenic variants in ITGB1BP1 are associated with cerebral cavernous malformations and immunodeficiency 49, highlighting its critical roles in maintaining vascular integrity and supporting immune system function. |
