| 背景信息 | Checkpoint kinase 1 (CHK1) is a conserved serine/threonine kinase that serves as a master regulator of the DNA damage response and cell cycle checkpoints. Upon the detection of DNA damage or replication stress, CHK1 is activated via phosphorylation by the upstream kinase ATR. Once active, CHK1 phosphorylates and promotes the degradation of CDC25 phosphatases, thereby inhibiting CDK1 and CDK2 to enforce intra-S and G2/M phase arrest. This delay allows for DNA repair and maintains genomic stability by preventing the entry of cells with damaged DNA into mitosis. CHK1 also stabilizes stalled replication forks and regulates the activity of WEE1 kinase. Due to its essential role in survival following DNA damage, CHK1 is a major therapeutic target in oncology, where its inhibition is used to sensitize cancer cells to DNA-damaging agents. |
