| 背景信息 | Complexin-1 (CPLX1) is a cytosolic protein that regulates synaptic vesicle exocytosis at presynaptic terminals. It binds to the assembled SNARE complex, clamping it in a fusion-competent state to prevent premature neurotransmitter release while priming vesicles for rapid, synchronous fusion upon calcium influx. This "fusion clamp" mechanism is vital for precise temporal control of synaptic transmission and motor control. Dysregulation of CPLX1 compromises synaptic efficacy. Clinically, mutations in CPLX1 are associated with severe neurodevelopmental disorders, including Developmental and Epileptic Encephalopathy 63 and Familial Infantile Myoclonic Epilepsy, manifesting as early-onset seizures and developmental delay due to impaired synaptic function. |
