| 背景信息 | Calpain 9 (CAPN9), also known as nCL-4, is a calcium-dependent cysteine protease that forms a heterodimer with the small regulatory subunit CAPNS1. Predominantly expressed in the gastrointestinal tract, particularly the stomach and small intestine, CAPN9 functions as part of the G-calpain complex to protect the gastric mucosa against injury through cytoskeletal remodeling and regulated proteolysis. Its activity is governed by unique autoinhibitory mechanisms that are released upon calcium binding. Beyond mucosal defense, CAPN9 participates in cell differentiation and TGF-beta-induced mesenchymal transitions. In oncology, downregulation of CAPN9 is strongly correlated with gastric cancer progression, advanced tumor stage, and poor prognosis, suggesting a tumor-suppressive role in maintaining gastric epithelial integrity. |
