| 背景信息 | T-box transcription factor 3 (T Box Protein 3) is a member of the T-box family characterized by a conserved T-domain that facilitates sequence-specific DNA binding. It functions primarily as a transcriptional repressor of genes such as p14ARF, p21WAF1, and PTEN, though it can also act as an activator in specific contexts. TBX3 is essential for the development of the heart, mammary glands, and limbs, and it maintains embryonic stem cell self-renewal by repressing differentiation-inducing genes. In the cardiac conduction system, it governs the identity of pacemaker cells by suppressing chamber myocardial programs. Clinically, haploinsufficiency of TBX3 causes Ulnar-mammary syndrome, a condition marked by limb, mammary, and cardiac defects. Additionally, TBX3 is frequently overexpressed in various cancers, where it promotes oncogenesis by enhancing cell proliferation, survival, and metastasis while contributing to drug resistance. |
