| 背景信息 | Axin 1 (AXIN1) is a cytoplasmic scaffold protein that serves as a core component of the β-catenin destruction complex, providing essential negative regulation of the canonical Wnt signaling pathway. Through its RGS and DIX domains, AXIN1 coordinates interactions between APC, GSK3B, CSNK1, and DVL to promote the phosphorylation and proteasomal degradation of β-catenin. Beyond Wnt signaling, it participates in TGF-β signaling and forms complexes with HIPK2 and p53 to regulate cell growth and apoptosis. Somatic and germline alterations in AXIN1 are well-established drivers of various malignancies, including hepatocellular carcinoma, hepatoblastoma, and medulloblastoma, as well as developmental anomalies such as caudal duplication, primarily due to the loss of Wnt pathway control. |
