| 背景信息 | ATP binding cassette subfamily C member 3 (ABCC3), also known as multidrug resistance‑associated protein 3 (MRP3), is an ATP‑dependent efflux transporter of the MRP subfamily that exports organic anions, including glucuronide and glutathione conjugates such as bilirubin diglucuronide, estradiol‑17β‑glucuronide and leukotriene C4, across the basolateral membrane of cells to protect tissues from bile acids, drugs, and toxic metabolites. Structurally, ABCC3 is a large membrane glycoprotein of about 1527 amino acids comprising three transmembrane domains that form the substrate translocation pathway and two conserved nucleotide‑binding domains that bind and hydrolyze ATP to power transport. ABCC3 is expressed mainly in liver, pancreas and adrenal glands, with lower levels in colon, kidney, lung and various epithelia, contributing to drug disposition and multi‑drug resistance by lowering intracellular accumulation of anticancer agents such as methotrexate and etoposide. Clinically, ABCC3 overexpression has been associated with chemoresistance and poor prognosis in cancers including non‑small cell lung cancer, pancreatic cancer and glioma, and variants have been linked to cholestatic conditions such as Dubin‑Johnson syndrome. |
