| 背景信息 | Serine peptidase inhibitor, Kunitz type 1 (SPINT1) is a transmembrane serine protease inhibitor that critically regulates the activation of Hepatocyte Growth Factor (HGF) by inhibiting HGF activator (HGFAC). It also targets other proteases such as matriptase to maintain epithelial homeostasis. SPINT1 is essential for preventing unrestricted proteolysis in tissues like the intestine and placenta. Pathogenic loss-of-function mutations cause Congenital Tufting Enteropathy (Diarrhea 5), a severe intestinal failure disorder characterized by intractable diarrhea and villous atrophy. Additionally, SPINT1 dysregulation is linked to placental insufficiency and altered MET signaling in cancer. |
