| 背景信息 | Fibroblast growth factor 16 (FGF16) is a secreted, heparin-binding growth factor that plays a significant role in embryonic development, cell proliferation, and tissue repair. It features a conserved beta-trefoil core domain that mediates binding to fibroblast growth factor receptors and heparan sulfate. Unlike many secreted proteins, FGF16 utilizes an internal hydrophobic motif for unconventional endoplasmic reticulum targeting and secretion. It signals primarily through FGFR1 and FGFR2 to regulate cardiac development and homeostasis. Clinically, pathogenic loss-of-function variants in the FGF16 gene are the cause of X-linked metacarpal 4-5 fusion, a congenital limb malformation. Emerging research also suggests its involvement in cardiac injury responses and metabolic regulation. |
