| 背景信息 | Guided entry of tail-anchored proteins factor 3, ATPase (GET3), also known as TRC40, is an ATPase essential for the post-translational insertion of tail-anchored (TA) proteins into the endoplasmic reticulum membrane. It features a nucleotide-binding domain and a zinc-coordinated homodimer interface that selectively captures cytosolic TA proteins. GET3 functions as a specific chaperone, shuttling substrates to the Get1/Get2 receptor complex at the ER for membrane integration. Beyond its targeting role, GET3 acts as a redox-regulated chaperone; under oxidative stress, it undergoes conformational changes involving zinc release and disulfide bond formation to prevent protein aggregation. Its function is critical for maintaining cellular proteostasis and organelle integrity. |
