| 背景信息 | Cytohesin 3 (CYTH3), also known as GRP1, is a guanine-nucleotide exchange factor (GEF) of the cytohesin family that activates ARF1 and ARF6 GTPases. It contains an N-terminal coiled-coil for homodimerization, a central catalytic Sec7 domain, and a C-terminal pleckstrin homology (PH) domain that mediates membrane recruitment via phosphatidylinositol-3,4,5-trisphosphate binding. CYTH3 regulates critical membrane trafficking events, including Golgi-to-ER retrograde transport, vesicle sorting, and epithelial polarization. Through its modulation of ARF signaling, it maintains Golgi structure and function. While specific monogenic disease associations are not well-defined, CYTH3 is integral to the cellular machinery governing vesicular transport and cytoskeletal organization. |
