| 背景信息 | Sperm associated antigen 9 (SPAG9), also known as JIP4, is a scaffolding protein that orchestrates MAPK signaling and microtubule-based transport. It specifically binds JNK2 and JNK3 to mediate c-Jun N-terminal kinase signaling and interacts with kinesin-1 and the dynein-dynactin complex to regulate organelle trafficking. Predominantly expressed in testicular germ cells and localized to the sperm acrosome, SPAG9 is essential for sperm-oocyte interaction and fertility. In pathological contexts, SPAG9 is aberrantly overexpressed in various malignancies, including prostate and lung cancers, where it promotes tumor growth and invasion via JNK pathway activation. It is also linked to retinitis pigmentosa phenotypes. |
