| 背景信息 | Ecotropic viral integration site 5 (EVI5) is a GTPase-activating protein (GAP) that primarily regulates Rab11, a small GTPase involved in vesicular trafficking, endocytosis, and cytokinesis. It contains a conserved N-terminal TBC domain that facilitates the hydrolysis of GTP to GDP on Rab11, thereby controlling its transition between active and inactive states. EVI5 also stabilizes FBXO5 to promote the accumulation of cyclin-A during interphase, which is essential for proper cell cycle progression. The protein localizes to centrosomes during interphase and relocalizes to the midbody during late mitosis to ensure successful cell division. Pathologically, the disruption of EVI5 leads to cytokinesis failure and the formation of multinucleate cells. Originally identified as a potential oncogene, EVI5 is critical for maintaining genomic stability and coordinating collective cell migration through Rab11-dependent pathways. |
