| 背景信息 | Gremlin 1 (GREM1) is a secreted glycoprotein and antagonist of the BMP signaling pathway, belonging to the DAN family. Characterized by a cysteine-knot motif, it binds and neutralizes BMP2 and BMP4, thereby modulating TGF-beta superfamily signaling. GREM1 is critical for embryonic development, particularly in limb bud formation, kidney morphogenesis, and angiogenesis. In adult tissues, aberrant GREM1 expression drives pathological processes; it promotes epithelial-to-mesenchymal transition (EMT), enhances invasiveness, and mimics VEGF signaling to support tumor angiogenesis. Elevated levels are associated with poor prognosis in cancers such as mesothelioma and colorectal carcinoma, where it fosters a metastatic microenvironment. |
