| 背景信息 | UDP-glucose 6-dehydrogenase (UGDH) is a cytosolic, homohexameric oxidoreductase that catalyzes the NAD+-dependent oxidation of UDP-glucose to UDP-α-D-glucuronate. This product is the essential precursor for glucuronidation and the biosynthesis of glycosaminoglycans, including hyaluronan, chondroitin sulfate, and heparan sulfate. These glycans are vital components of the extracellular matrix, supporting embryonic development, cell migration, and tissue repair. Clinically, inherited loss-of-function variants in UGDH are linked to early-onset epileptic encephalopathy, highlighting its role in neurodevelopment. In oncology, UGDH-driven overproduction of hyaluronan is associated with tumor progression, metastasis, and poor prognosis in various epithelial cancers. Consequently, UGDH serves as both a molecular marker and a potential therapeutic target for inhibiting the invasive properties of cancer cells and modulating the tumor microenvironment. |
