| 背景信息 | ADAM metallopeptidase with thrombospondin type 1 motif 4 (ADAMTS4) is a member of the ADAMTS protein family, characterized by a multi-domain structure including a propeptide region, a metalloproteinase domain, a disintegrin-like domain, and a thrombospondin type 1 motif. This enzyme is primarily known for its role in the degradation of aggrecan, a major proteoglycan of cartilage, and brevican, a brain-specific extracellular matrix protein. The full-length ADAMTS4 (70 kDa) is the most effective aggrecanase, but it exhibits little activity against the Glu(373)-Ala(374) bond, a signature site of aggrecanase activity. ADAMTS4 is involved in the turnover of aggrecan and may play a critical role in the destruction of aggrecan in arthritic diseases and potentially in neurodegenerative conditions such as Alzheimer's disease. Its expression is upregulated in arthritic disease, contributing to disease progression through the degradation of aggrecan. |
