| 背景信息 | ZPR1 zinc finger (ZPR1) is a highly conserved eukaryotic chaperone protein containing two tandem zinc-binding domains. In quiescent cells, it is primarily cytoplasmic and interacts with the kinase domain of the epidermal growth factor receptor (EGFR). Upon mitogen stimulation, ZPR1 dissociates from EGFR and translocates to the nucleolus, where it supports preribosomal RNA processing and cell cycle progression. ZPR1 also functions as a critical regulator of translation elongation by binding to eEF1A, stabilizing its GTP-bound conformation and protecting it from degradation. This interaction is essential for preventing proteotoxic stress and maintaining protein synthesis. Furthermore, ZPR1 interacts with the survival motor neuron (SMN1) protein to facilitate pre-mRNA splicing and promote neuronal differentiation. Depletion of ZPR1 leads to eEF1A insufficiency, protein aggregation, and growth arrest, highlighting its importance in cellular proteostasis and development. |
