| 背景信息 | Programmed cell death 6 (PDCD6), also known as ALG-2, is a penta-EF-hand calcium-binding protein that functions as a calcium sensor in intracellular trafficking and signaling. It undergoes conformational changes upon calcium binding to interact with diverse targets, bridging ESCRT complexes and regulating COPII-mediated ER-to-Golgi transport, endosomal biogenesis, and membrane repair. While originally linked to apoptosis, PDCD6 also functions as a negative regulator of angiogenesis by inhibiting the VEGFR2-PI3K-Akt pathway. Dysregulation of PDCD6 is observed in various malignancies; it is associated with ocular melanoma and hereditary Wilms' tumor, and its expression correlates with progression in gastric cancer. |
