| 背景信息 | MacroH2A.1 histone (MACROH2A1) is a histone H2A variant characterized by a unique C-terminal macrodomain connected to the histone fold. It is enriched in facultative heterochromatin, including the inactive X chromosome, where it stabilizes nucleosomes and recruits HDAC1/2 to mediate transcriptional repression and chromatin compaction. MacroH2A1 regulates genomic stability by modulating replication fork progression and coordinating the loading of the MCM helicase complex. Through alternative splicing, it produces isoforms that differentially influence cell plasticity and metabolism. MacroH2A1 acts as a barrier to cellular reprogramming and its dysregulation is implicated in cancer progression and metabolic gene control. |
