| 背景信息 | H2A.P histone (H2AP) is a 37-amino acid N-terminal peptide fragment derived from histone H2A, a core eukaryotic histone responsible for packaging DNA into chromatin. This peptide features a clustered DNA-binding region with an alpha-helical structure that is essential for electrostatically driven DNA condensation and nuclear import via its nuclear localization signal. Unlike full nucleosomes, this fragment enables efficient gene transfection in cell lines without the requirement for H2B. Canonical H2A contains a conserved histone fold and terminal tails that undergo post-translational modifications, such as ubiquitination, to regulate DNA repair through interactions with BRCA1/BARD1 and RNF168. Additionally, H2A variants like H2A.Z and macroH2A modulate chromatin dynamics, transcription, and genomic stability by altering the acidic patch and linker DNA protection. |
