| 背景信息 | Nuclear receptor subfamily 1 group I member 2 (NR1I2) is a ligand-activated nuclear receptor and DNA-binding transcription factor that serves as a master regulator of xenobiotic detoxification. Characterized by a C4 zinc-finger DNA-binding domain and a flexible ligand-binding domain, it accommodates a wide range of structurally diverse xenobiotics and endogenous steroids. Upon activation, NR1I2 forms heterodimers with the retinoid X receptor (RXR) to bind response elements in the promoters of target genes, notably inducing the expression of phase I, II, and III drug-metabolizing enzymes and transporters, such as CYP3A4 and ABCB1/MDR1. Predominantly expressed in the liver and intestine, it plays a critical role in mediating drug-drug interactions and maintaining bile acid, cholesterol, and vitamin D homeostasis. Clinically, dysregulation of NR1I2 is associated with altered drug responses and metabolic conditions such as cerebrotendinous xanthomatosis and osteomalacia. |
