| 背景信息 | Leucine-rich repeat containing G protein-coupled receptor 5 (LGR5) is a seven-transmembrane G protein-coupled receptor characterized by a large N-terminal extracellular domain containing 17 leucine-rich repeats. It functions as a receptor for R-spondins, potentiating canonical Wnt/beta-catenin signaling by inhibiting the transmembrane E3 ligases RNF43 and ZNRF3, thereby stabilizing Frizzled receptors. LGR5 undergoes constitutive internalization and is a marker of adult stem cells in the intestine, stomach, and hair follicles, identifying cycling multipotent progenitors essential for tissue renewal. Aberrant upregulation of LGR5 is observed in colorectal and gastric cancers, where it drives tumorigenesis by maintaining stem cell properties. Consequently, LGR5 is a critical target for investigating stem cell biology and cancer therapeutics. |
