| 背景信息 | PC4 and SRSF1 interacting protein 1 (PSIP1), also known as LEDGF/p75, is a chromatin-associated protein that functions as a transcriptional coactivator and epigenetic reader. Through its PWWP domain, it binds H3K36-methylated nucleosomes to tether protein complexes to transcriptionally active chromatin. Notably, PSIP1 recruits HIV-1 integrase to host chromatin, facilitating viral integration into active genes. It also plays a key role in stress responses as a cofactor for HSF1. In pathology, PSIP1 is implicated in MLL-rearranged leukemias, where it stabilizes oncogenic fusion proteins, and contributes to cancer progression and therapy resistance through transcriptional reprogramming. |
