| 背景信息 | TNF receptor superfamily member 21 (TNFRSF21), also known as Death Receptor 6 (DR6), is a type I transmembrane protein belonging to the TNF receptor superfamily. It is primarily expressed in the thymus, spleen, and white blood cells, and plays a crucial role in apoptosis and immune regulation. The protein consists of a 308 amino acid extracellular domain with four cysteine-rich motifs, a 21 amino acid transmembrane segment, and a 285 amino acid cytoplasmic region containing one death domain. DR6 is involved in the activation of NF-κB and MAPK8/JNK pathways, leading to cell apoptosis. It also interacts with TRADD protein, which mediates signal transduction of TNF-receptors. DR6 is implicated in various diseases, including Alzheimer's disease, where it is involved in neurodegeneration and axonal pruning, and in cancer, where it can induce endothelial cell necroptosis allowing tumor metastasis. Elevated serum levels of DR6 are associated with late-stage ovarian cancer. |
