| 背景信息 | Trio Rho guanine nucleotide exchange factor (TRIO) is a large, multidomain protein that serves as a critical regulator of the actin cytoskeleton, cell migration, and adhesion. It functions as a dual Rho family GEF, utilizing two distinct catalytic units: the N-terminal TrioD1 domain activates Rac1 and RhoG, while the C-terminal TrioD2 domain activates RhoA. This coordinated regulation is essential for neuronal development, axon guidance, and leukocyte transendothelial migration. TRIO also contains spectrin repeats and a serine/threonine kinase domain, supporting its role as a versatile signaling scaffold. Clinically, mutations and dysregulation of TRIO are implicated in the progression of various malignancies, particularly G-alpha-q/11-driven cancers, where the relief of autoinhibition in its RhoA-activating module sustains oncogenic signaling and promotes tumor growth. |
