| 背景信息 | Dimethylarginine dimethylaminohydrolase 1 (DDAH1) is a cytosolic enzyme essential for maintaining vascular homeostasis through the regulation of nitric oxide (NO) signaling. It functions by hydrolyzing asymmetric dimethyl-L-arginine (ADMA) and Nω-monomethyl-L-arginine (MMA), which are endogenous inhibitors of nitric oxide synthase (NOS). By clearing these inhibitors, DDAH1 facilitates the continuous production of NO. Structurally, the protein belongs to the DDAH gene family and forms a characteristic propeller-like fold. Its catalytic mechanism involves a conserved cysteine residue within the active site, which is susceptible to inhibition by endogenous zinc ions (Zn2+). This enzymatic activity is a critical determinant of systemic NO bioavailability and vascular tone. |
